Mechanism & Pharmacokinetics ▼
Mechanism: Amlodipine relaxes vascular smooth muscle, reducing peripheral resistance. Olmesartan blocks angiotensin II type 1 receptors, reducing vasoconstriction and aldosterone secretion.
Pharmacokinetics: Amlodipine: oral Tmax 6–12 hr, metabolized in liver. Olmesartan: oral Tmax 1–2 hr, metabolized to active form, excreted in urine and feces; half-life ~13 hr.
