Mechanism: Tropicamide is a muscarinic antagonist that causes cycloplegia and pupil dilation. Phenylephrine is an α1-adrenergic agonist that stimulates the dilator pupillae muscle, producing mydriasis. Combined, they achieve rapid and sustained pupil dilation for examination.
Pharmacokinetics: Topically applied; minimal systemic absorption. Onset 15–30 minutes; duration 4–6 hours. Metabolized minimally; excreted mainly via urine.
