Mechanism & Pharmacokinetics ▼
Mechanism: Paracetamol inhibits central prostaglandin synthesis providing analgesic and antipyretic effects. Caffeine acts as a central nervous system stimulant and enhances analgesic effect of paracetamol.
Pharmacokinetics: Rapid oral absorption. Paracetamol metabolized via glucuronidation and sulfation in liver. Caffeine metabolized by CYP1A2; excreted mainly in urine. Half-life: Paracetamol 2–3 h, Caffeine 3–7 h.
