Mechanism: Nucleotide analog inhibitor of HCV NS5B RNA-dependent RNA polymerase. Incorporation into viral RNA leads to chain termination and inhibits viral replication.
Pharmacokinetics: Rapidly absorbed; peak plasma concentration in ~0.5–2 hours. Metabolized to active triphosphate GS-461203. Mainly eliminated via urine. Half-life of active metabolite ~27 hours.
