Common Dose80–125 mg
PregnancyConsidered Safe
Indications ▼
Relief of abdominal bloating
Excessive gas accumulation
Post-operative gas discomfort
Infantile colic
Adjunct in diagnostic procedures (endoscopy, radiography)
Mechanism & Pharmacokinetics ▼
Mechanism: Simethicone reduces surface tension of gas bubbles in the gastrointestinal tract, causing them to coalesce and be eliminated more easily through belching or passage of flatus. It has no effect on gastric acid secretion or motility.
Pharmacokinetics: Simethicone is physiologically inert. It is not absorbed from the gastrointestinal tract, not metabolized, and excreted unchanged in feces.

Clinical / Research Summary ▼
Therapeutic Role: Simethicone is an anti-foaming agent used to relieve bloating, discomfort, and gas in the gastrointestinal tract. It is commonly used in functional dyspepsia, irritable bowel syndrome (IBS), and post-operative gas relief.
Mechanism-Based Evidence: It reduces surface tension of gas bubbles in the stomach and intestines, causing them to coalesce and be expelled more easily through belching or flatulence. It is not systemically absorbed, making systemic side effects rare.
Clinical Effectiveness: Clinical studies support its use for symptomatic relief of bloating and postprandial gas. Often combined with antacids or lactase preparations in gastrointestinal discomfort management.
Safety Profile: Considered very safe with minimal adverse effects; occasional mild gastrointestinal upset may occur. Safe in pregnancy and lactation due to lack of systemic absorption.
Combination Therapy: Frequently co-administered with antacids (e.g., magnesium hydroxide/aluminium hydroxide) for dual relief of heartburn and bloating.
Current Research Directions: Focus on its use in pediatric colic, postoperative abdominal discomfort, and as adjunct therapy in functional gastrointestinal disorders.
