Mechanism & Pharmacokinetics ▼
Mechanism: Acts as a cofactor for gamma-glutamyl carboxylase, enabling synthesis of clotting factors II, VII, IX, and X, as well as proteins C and S, restoring normal coagulation.
Pharmacokinetics: Rapidly absorbed and distributed after IM or IV injection. Metabolized in liver and excreted mainly via bile. Onset: IM 1–2 hours; IV 15–60 min. Duration: 12–24 hours.

Rare: Anaphylactoid reactions, hypersensitivity, hyperbilirubinemia in neonates if IV high dose

Hypersensitivity to phytomenadione or any component of the formulation
Severe anaphylactic reaction history to vitamin K injections

Warfarin or other coumarin anticoagulants: may reduce anticoagulant effect
Bile acid sequestrants: may reduce absorption
Orlistat: may decrease vitamin K absorption
